The objective of this study was to evaluate the use of luteal color doppler (CD) ultrasonography and plasma concentrations of pregnancy-associated glycoproteins (PAG) for early pregnancy diagnosis in Bos taurus beef cows. Additionally, CD and PAG were evaluated as potential predictors of late embryonic/early fetal mortality (LEM). Postpartum beef cows (n = 212) were exposed to estrus synchronization followed by fixed-time artificial insemination (day 0). On days 20 and 22, B-mode and CD ultrasonography were performed to evaluate corpus luteum (CL) morphometries and blood perfusion. Moreover, blood samples were collected on days 25 and 29 to quantify circulating concentrations of PAG. Conventional ultrasonography on days 29 and 100 was utilized as the gold-standard method for pregnancy diagnosis. Cows that experienced pregnancy loss between days 29 and 100 were classified as LEM. Pregnant cows had larger and more vascularized CL compared with nonpregnant cows on days 20 and 22 (P < 0.001 for all response variables). Accuracy for CD on days 20 and 22 were 87% and 92%, respectively. Accuracy for PAG on days 25 and 29 were 84% and 99%, respectively. No false negative (FN) results were observed for CD on both days 20 and 22; however, there were 7.1% FN results for PAG on day 25. Cows that experienced LEM had decreased (P = 0.04) circulating PAG on day 29 of gestation compared with cows that maintained pregnancy; however, there were no differences in luteal blood perfusion on days 20 and 22 (P ≥ 0.53) or circulating PAG on day 25 (P = 0.46) between LEM cows and cows that maintained pregnancy. Sensitivity and specificity of PAG on day 29 as predictors of LEM were 83% and 77%, respectively. In conclusion, CD resulted in accurate pregnancy diagnosis in B. taurus beef cows on both days 20 and 22 of gestation, while having no FN results. Circulating concentrations of PAG were decreased in cows that experienced LEM; however, further research is required to utilize PAG as a predictor of LEM commercially.


Early pregnancy detection allows cattle producers to increase reproductive efficiency. Pregnancy failures associated with embryonic mortality represents an economic burden to the beef production industry. The present study evaluated the use of color doppler (CD) ultrasonography of specific ovarian structures and pregnancy-associated glycoproteins (PAG) in plasma as potential alternatives to diagnose pregnancy earlier than industry-standard methods in beef cows with Bos taurus genetics. Additionally, the present study evaluated the use of these methods to predict embryonic mortality during early gestation. The results of the present study indicate that: 1) CD can accurately recognize most nonpregnant cows with B. taurus genetics as early as 20 days after breeding. 2) Plasma concentrations of PAG resulted in suboptimal accuracy on day 25 of gestation. 3) Cows that undergo pregnancy loss between 29 and 100 d after breeding have decreased concentrations of PAG in their circulation on day 29, even though they were identify as pregnant with ultrasonography on the same day. 4) Both blood concentrations of PAG and luteal blood perfusion estimated by CD failed to accurately predict pregnancy loss.

To determine the effect of CaO-treated Pensacola bahiagrass (Paspalum notatum) hay on intake, ruminal fermentation parameters, and apparent total-tract digestibility of nutrients, nine ruminally cannulated Angus-crossbred steers were used in a triplicated 3 × 3 Latin square design. Steers had ad libitum access to either 1) untreated dry hay (DH; n = 8); 2) hay at 50% DM treated with 8.9% CaCO3 (dry matter [DM] basis; CC; n = 9); or 3) hay at 50% DM treated with 5% CaO (DM basis; CO; n = 8). Water was added to reach 50% DM in the CC and CO diets. Ruminal fluid and blood samples were collected every 3 h for 24 h. Ruminal fluid was analyzed for pH, volatile fatty acids (VFA), and ammonia-nitrogen (NH3-N). Blood was analyzed for plasma urea nitrogen (PUN). Hay and fecal samples were collected for 4 d, four times daily for hay and twice daily for feces, to determine apparent total-tract digestibility of nutrients. The hay provided to steers during the digestibility period was analyzed for in vitro organic matter digestibility (IVOMD) for 48 h. Data were analyzed as repeated measures for blood and ruminal fermentation parameters. Total DM intake was not affected (P ≥ 0.674) by treatment. A treatment effect (P < 0.001) was observed for average ruminal pH, where steers consuming CO had the greatest pH (P < 0.001). Ruminal concentration of NH3-N tended (P = 0.059) to be reduced in steers consuming CO. There was a treatment × time interaction (P = 0.023) on concentrations of PUN, where at 3 h DH and CO were lesser than CC (P ≤ 0.050) and at 21 h DH was lesser than CC (P = 0.020). Total VFA, acetate, propionate, butyrate, branched-chain VFA, and valerate concentrations were affected by treatment (P ≤ 0.035), where a reduction (P ≤ 0.034) occurred in steers consuming CO. No treatment differences were observed for total-tract digestibility of DM (P = 0.186), organic matter (P = 0.169), or crude protein (P = 0.152); however, steers consuming DH had greater neutral detergent fiber (P = 0.038) than CC and tended to be greater than CO (P = 0.082). The CO hay had greater (P = 0.005) IVOMD compared with DH and tended (P = 0.100) to be greater than CC. Bahiagrass hay treated with CaO may reduce ruminal fermentation, as indicated by decreased total VFA concentration without altering DM intake. The addition of CaO did not improve the digestibility of bahiagrass hay in vivo; however, in vitro results are contradictory and warrant further elucidation.


With the ever-growing desire to increase efficiency in beef cattle production, researchers have developed strategies such as treating poor-quality forages with chemicals to increase the digestibility of fiber fractions, consequently increasing their energy value for cattle feeding. Calcium oxide has been proposed as a replacement to more caustic chemicals used in the past (e.g., NaOH) and data indicate that it can promote similar and effective outcomes. The current study evaluated the effects of bahiagrass hay treated with calcium oxide on ruminal fermentation parameters, apparent total-tract digestibility of nutrients, and intake by beef steers consuming hay ad libitum as the sole ingredient in their diet. Additionally, in vitro organic matter digestibility was evaluated on the hay provided to steers to assess treatment effectiveness. Results indicated that steers consuming bahiagrass hay treated with calcium oxide had 1) increased pH and reduced volatile fatty acids concentrations in the rumen; 2) reduced or tendency for reduction on total-tract digestibility of fiber fractions; and 3) no effect on intake, all when compared with steers consuming untreated hay. In contrast, in vitro results indicated that organic matter digestibility was increased when the forage was treated with calcium oxide.

To determine effects of delaying the injection of prostaglandin F2α (PGF) and fixed-time artificial insemination (TAI) in the 14-d CIDR-PG protocol, 1,049 Angus heifers at six locations were enrolled in a completely randomized design. Within location heifers were randomly assigned to one of two treatment groups: 1) PG16 (n = 518), heifers received a controlled internal drug release (CIDR) insert on d 0 for 14 d, a 25-mg injection of PGF 16 d after CIDR removal (d 30), and a 100-µg injection of gonadotropin-releasing hormone concurrent with TAI 66 ± 2 h later; or 2) PG17 (n = 531), heifers were treated the same as PG16, however, PGF was administered 17 d after CIDR removal (d 31), and heifers were TAI 66 ± 2 h later. Estrus detection patches were applied to a subset (n = 482) of heifers at the time of PGF administration and were examined for activation at TAI. Dominant follicle diameter was determined via transrectal ultrasonography at PGF administration and TAI in a subset of heifers (n = 116). Transrectal ultrasonography was performed to determine pregnancy rates to TAI (PR/AI) between 30 and 45 d after TAI. Estrus expression prior to TAI differed by treatment where PG17 heifers had greater (P < 0.01) expression of estrus than PG16 heifers (57.8 ± 6.1% vs. 43.4 ± 6.1%, respectively). Nevertheless, dominant follicle diameters at PGF and at TAI were similar (P ≥ 0.59) between PG16 and PG17 heifers. In addition, PR/AI did not differ (P = 0.29) between PG16 and PG17 treatments (50.5 ± 3.2% vs. 45.7 ± 3.1%, respectively). Results of this experiment indicate that delaying the injection of PGF and TAI in the 14-d CIDR-PG protocol increased estrus expression prior to TAI yet did not improve fertility in beef heifers.

The aim of this study was to evaluate the effect of bovine somatotropin (bST) on fetal and placental development during the first third of gestation in beef heifers. Angus heifers (n = 97) were randomly assigned to either receive a 500-mg injection of bST (BST) biweekly on days 0, 15, 29, 43, and 57 of gestation or not receive bST (CTL) throughout the experiment. Body weight (BW) was assessed on days -9, -3, 0, 15, 22, 29, 43, 50, 57, 64, and 77, while blood samples were collected on days 0, 22, 50, and 64. Pregnancy status was determined via transrectal ultrasonography on days 29 and 64. A subset of pregnant heifers (BST, n = 7; CTL, n = 5) were harvested on day 84, and complete gravid reproductive tracts and liver tissue were collected for analysis. Cytochrome P450 1A (CYP1A), 2C (CYP2C), 3A (CYP3A), and uridine 5'-diphospho-glucuronosyltransferase (UGT) activities were determined. Mean change in BW and average daily gain of heifers between fixed-time artificial insemination (day 0) and day 77 did not differ between treatments (P ≥ 0.05). Mean concentrations of insulin-like growth factor 1 (IGF-1) were greater (P < 0.001) in BST (347 ± 27.7 ng/mL) compared with CTL (135 ± 32.8 ng/mL) heifers. Mean placental weight, fetal membrane weight, uterine weight, and ovarian and corpus luteum (CL) weights, as well as fetal morphometric data, did not differ (P ≥ 0.05) between treatments. However, BST heifers had greater (P = 0.03) quantities of combined fetal fluid compared with CTL (521.6 ± 22.9 vs. 429.6 ± 27.14 g, respectively). Tendencies were observed for BST heifers to have reproductive tracts with fewer placentomes (P = 0.08) and fetuses with greater umbilical diameters (P = 0.09) compared with CTL. The activity of CYP1A did not differ (P ≥ 0.05) within the maternal and fetal liver, caruncle, cotyledon, or CL tissue samples between treatments. Furthermore, CYP3A activity was only observed in maternal liver samples and was not different between treatments (P ≥ 0.05). Interestingly, CYP2C activity was greater (P = 0.01) in the liver of BST vs. CTL heifers, and UGT activity was greater (P = 0.02) in the CL from BST heifers compared with CTL. In conclusion, the administration of bST during the first third of gestation increased plasma concentrations of IGF-1, which resulted in an increase in fetal fluid, decrease in placentome number, and greater umbilical diameter, but failed to alter fetal development.

The objective of this experiment was to investigate the relationship between residual feed intake (RFI) and parameters associated with reproductive efficiency in growing beef heifers. One hundred and seventy-nine replacement beef heifers (Bos taurus and Bos indicus-influenced) were enrolled in a retrospective cohort design. Heifers were assigned to a 70-d feed efficiency test, and results were utilized to retrospectively classify heifers into low (n = 51), medium (n = 66), or high (n = 62) RFI groups based on their individual RFI values. Blood samples were collected weekly throughout the feed efficiency test and breeding season, which were analyzed for plasma concentration of progesterone to determine age at puberty (PUB). By design, residual feed intake differed among RFI groups where high (1.21 ± 0.08 kg/d) had greater (P < 0.01) RFI than medium (0.00 ± 0.08 kg/d) and low RFI groups (-1.49 ± 0.09 kg/d), and medium had greater (P < 0.01) RFI than low RFI heifers. Yet, no differences were observed between breed types for RFI (P = 0.53). Average daily gain (ADG) differed between breed types where Bos taurus heifers had greater ADG (P = 0.02) than B. indicus-influenced heifers. Furthermore, ADG tended (P = 0.08) to differ among RFI groups, where medium RFI heifers tended to have reduced ADG compared with low and high RFI heifers. No differences were determined between breed types for gain-to-feed ratio (G:F; P = 0.20; however, G:F differed among RFI groups and was greater (P < 0.01) in low vs. high, and low vs. medium RFI heifers. The percentage of cycling heifers by the start of the breeding season differed (P = 0.03) by RFI group, where a greater percentage of low RFI heifers were cycling compared with high RFI heifers. A difference was determined on PUB between breed types (P = 0.03), where Bos taurus (393.40 ± 4.64 d) heifers had a reduced PUB compared with B. indicus-influenced (406.90 ± 5.07 d) heifers. In addition, PUB differed by RFI group where low (385.96 ± 6.20 d; P < 0.01) and medium (398.47 ± 5.47 d; P = 0.02) RFI heifers had a reduced PUB compared with High RFI heifers (416.03 ± 5.58 d). In conclusion, more feed efficient heifers attained PUB earlier than less feed efficient heifers. Therefore, selection of heifers for feed efficiency may positively impact reproductive performance of replacement beef heifers.

Two randomized block designs were performed to evaluate the effects of bismuth subsalicylate (BSS) and encapsulated calcium-ammonium nitrate (eCAN) on enteric methane production, nutrient digestibility, liver mineral concentration, and performance of beef cattle consuming bahiagrass hay (Paspalum notatum; ad libitum) and sugar cane molasses [1.07 kg/d; dry matter basis]. Experiment 1, used 25 crossbred steers [335 ± 46 kg of initial body weight (BW)] with a 2 × 2 + 1 factorial arrangement of treatments for two 20 d periods. Factors were nonprotein nitrogen (NPN) source (350 mg/kg BW of nitrate or 182 mg/kg BW of urea), BSS (0 or 58.4 mg/kg BW), and a negative control (NCTRL; bahiagrass hay and molasses only). Steers were re-randomized for a second period (n = 10/treatment total). Intake, apparent total tract digestibility and enteric methane were evaluated. Experiment 2 used 75 crossbred heifers in 25 pens (3 heifers/pen; 279 ± 57 kg of initial BW), consuming the same diet and treatments as experiment 1, to determine liver mineral concentration and growth performance over 56 d. Orthogonal contrasts were used to evaluate the effects of NPN (NCTRL vs. others), source of NPN (NS; urea vs. eCAN), BSS, and NS × BSS. For experiment 1, no interactions were observed for any variables, nor were there any effects of NPN on total tract digestibility of nutrients, except for crude protein. Digestibility of all nutrients was reduced (P ≤ 0.021) for steers consuming eCAN compared with urea. There was no effect (P > 0.155) of BSS on digestibility of nutrients; however, BSS reduced (P = 0.003) apparent S retention. Enteric CH4 emission (g/kg BW0.75) was decreased (P = 0.051) by 11% with the addition of eCAN compared with urea. For experiment 2, no NS × BSS interactions (P ≥ 0.251) were observed to affect liver mineral concentration; however, the addition of BSS decreased liver concentration of Cu (P = 0.002) while increasing Fe concentration (P = 0.016). There was an NS × BSS interaction (P = 0.048) where heifers consuming eCAN and BSS had lesser final BW compared with heifers consuming urea and BSS. While eCAN may be a viable resource for mitigating enteric CH4 production of forage-fed cattle, the negative effects on digestibility should be considered. Furthermore, BSS, at the amount provided, appears to have no negative effects on digestibility of nutrients in forage-fed cattle; however, there may be deleterious impacts on performance depending upon what nitrogen source is supplied.

A replicated 5 × 5 Latin square design with a 2 × 2 + 1 factorial arrangement of treatments was used to determine the effects of bismuth subsalicylate (BSS) and encapsulated calcium ammonium nitrate (eCAN) on ruminal fermentation of beef cattle consuming bahiagrass hay (Paspalum notatum) and sugarcane molasses. Ten ruminally cannulated steers (n = 8; 461 ± 148 kg of body weight [BW]; average BW ± SD) and heifers (n = 2; 337 ± 74 kg of BW) were randomly assigned to one of five treatments as follows: 1) 2.7 g/kg of BW of molasses (NCTRL), 2) NCTRL + 182 mg/kg of BW of urea (U), 3) U + 58.4 mg/kg of BW of BSS (UB), 4) NCTRL + 538 mg/kg of BW of eCAN (NIT), and 5) NIT + 58.4 mg/kg of BW of BSS (NITB). With the exception of NCTRL, all treatments were isonitrogenous. Beginning on day 14 of each period, ruminal fluid was collected and rectal temperature was recorded 4× per day for 3 d to determine ruminal changes every 2 h from 0 to 22 h post-feeding. Ruminal gas cap samples were collected at 0, 3, 6, 9, and 12 h on day 0 of each period followed by 0 h on days 1, 2, 3, and 14. Microbial N flow was determined using Cr-Ethylenediaminetetraacetic acid, YbCl3, and indigestible neutral detergent fiber for liquid, small particle, and large particle phases, respectively. Data were analyzed using the MIXED procedure of SAS. Orthogonal contrasts were used to evaluate the effects of nonprotein nitrogen (NPN) inclusion, NPN source, BSS, and NPN source × BSS. There was no treatment effect (P > 0.05) on concentrations of H2S on day 0, 1, 2, or 14; however, on day 3, concentrations of H2S were reduced (P = 0.018) when NPN was provided. No effect of treatment (P = 0.864) occurred for ruminal pH. There was an effect of NPN source on total concentrations of VFA (P = 0.011), where a 6% reduction occurred when eCAN was provided. There were effects of NPN (P = 0.001) and NPN source (P = 0.009) on the concentration of NH3-N, where cattle consuming NPN had a greater concentration than those not consuming NPN, and eCAN reduced the concentration compared with urea. Total concentrations of VFA and NH3-N were not affected (P > 0.05) by BSS. There was an effect of BSS (P = 0.009) on rectal temperature, where cattle not consuming BSS had greater temperatures than those receiving BSS. No differences for NPN, NPN source, nor BSS (P > 0.05) were observed for microbial N flow. In conclusion, eCAN does not appear to deliver equivalent ruminal fermentation parameters compared with urea, and BSS has limited effects on fermentation.

This experiment was conducted to evaluate effects of estrous expression and intensity of behavioral estrous expression, assessed by evaluating physical activity, on variables associated with fertility in beef cows. The following treatment regimen was imposed on lactating, multiparous cows (n = 273): 100-µg injection of GnRH and an intravaginal progesterone (P4) releasing device (CIDR) administered on d -10, 25-mg injection of prostaglandin F2α at CIDR removal on d -3, and another GnRH injection + fixed-time artificial insemination (TAI) 60-66 h following CIDR removal (d 0). Cows were fitted with a pedometer on d -10, and an estrous detection patch on the tail-head on d -3. Cows were classified as not expressing estrus (NOESTR) or expressing estrus with net physical activity greater (HIESTR) or less than the median (LWESTR). Only data from cows responsive to the estrous synchronization treatment regimen were utilized (NOESTR, n = 119; LWESTR, n = 50; HIESTR, n = 50). Diameter of dominant follicles on d 0 and corpus luteum volume on d 7 were greater in HIESTR compared with LWESTR and NOESTR groups (P < 0.01). Plasma P4 concentrations on d 7 and pregnancy rates to TAI were greater in HIESTR and LWESTR than the NOESTR group (P < 0.01). Hence, if there was estrous expression during the period of the GnRH-based TAI treatment regimen, there were greater pregnancy rates to TAI, whereas when there was greater intensity of behavioral estrous symptoms, there were larger dominant follicles and corpora lutea dimensions but not greater pregnancy rates.

To evaluate how the inclusion of Bos indicus genotype influences early fetal development in cattle, a reciprocal embryo transfer approach was used in a completely randomized design with a 2 × 2 × 2 factorial arrangement of treatments to generate 55 pregnancies over 2 consecutive years (n = 55). Recipient cows were randomly assigned to (i) a diet that met daily energy maintenance requirements (MAINT) or (ii) a diet that restricted intake to 70% of the energy maintenance requirements (RESTR). Angus (AN) and Brangus (BN) embryo donors were superovulated and artificially inseminated with female sexed-sorted semen from the same breed. Embryos were then randomly transferred to either AN or BN recipients fed their respective diets for 28 d. Recipients remained on the dietary scheme until day 91 of gestation and were then comingled and fed a common diet that met their energy requirements until calving. Measurements included pregnancy establishment at day 28 of gestation, interferon-stimulated genes (ISG) expression in peripheral blood leukocytes, pregnancy-associated glycoproteins (PAG; using two commercial [A1 and A2] and one in-house assay), and fetal crown-to-rump length (CRL). Recipients in the RESTR diet had lower BWs and BCS (diet × day; P < 0.01) than MAINT recipients. Energy-restricted AN recipients experienced greater (recipient breed × diet, P < 0.01) pregnancy failure by day 28 than the other recipient breed × diet combinations. Restricted recipients that received AN embryos experienced greater pregnancy failure than RESTR recipients receiving BN embryos (embryo breed × diet; P = 0.03). No relevant differences were observed in ISG expression (P > 0.10). Recipients that received BN embryos had greater plasma concentrations of PAG in both A1 (embryo breed × day, P < 0.01) and A2 (embryo breed; P < 0.01). Alternatively, recipients that received AN embryos had greater plasma concentrations of PAG for the in-house assay (embryo breed × day; P < 0.01). In addition, fetuses from AN recipients had greater CRL on day 91 (breed × day, P < 0.01). In summary, Bos taurus cows experienced greater pregnancy failure when nutrient restricted. Furthermore, fetal size and the profile of PAG production during early gestation differed between B. indicus-influenced and B. taurus cattle.

Our objectives were to determine the effects of the administration of recombinant bovine somatotropin (bST) at the initiation of a fixed-time AI (TAI) protocol on concentrations of plasma IGF-1, follicle diameter, embryo/fetal size, and pregnancy rates in replacement beef heifers. Four hundred and fourteen Angus-based beef heifers were enrolled in a completely randomized design at 4 locations from January to July of 2016. All heifers were exposed to the 7-d CO-Synch + controlled internal drug release (CIDR) protocol where they received a 100-µg injection of GnRH and a CIDR insert on day -9, 25 mg of PGF2α at CIDR removal on day -2, followed by a 100-µg injection of GnRH and TAI 54 ± 2 h later on day 0. Within location, all heifers were randomly assigned to 1 of 2 treatments: 1) heifers that received 650 mg of bST on day -9 (BST; n = 191); or 2) heifers that did not receive bST on day -9 (CONTROL; n = 223). Blood samples were collected on day -9, 0, 28, and 60 to determine the plasma concentrations of IGF-1. Follicle diameter was determined on day -2 and 0 by transrectal ultrasonography. Pregnancy was diagnosed via transrectal ultrasonography on day 28 or 35, and again at least 30 d after the end of the breeding season. Embryo morphometry was assessed by measuring crown-to-rump length (CRL) on day 28, and fetal size was assessed by measuring crown-to-nose-length (CNL) on day 60. Concentrations of plasma IGF-1 did not differ between treatments on day -9 (P = 0.924), 28 (P = 0.075), and 60 (P = 0.792); however, concentrations of plasma IGF-1 were greater (P < 0.001) in BST-treated heifers at TAI (372.4 ± 16.6 vs. 193.7 ± 16.6 ng/ml). No differences (P = 0.191) were detected for follicle diameter between CONTROL and BST treatments on day -2 or 0. Pregnancy rates to TAI (PR/AI) were greater (P = 0.028) for CONTROL compared to BST heifers (42.5 ± 4.0 vs. 29.9 ± 4.1%). No differences (P = 0.536) in CRL were observed on day 28 between CONTROL and BST heifers. In addition, no difference (P = 0.890) was observed for CNL between CONTROL and BST treatments. Final pregnancy rates did not differ (P = 0.699) between treatments. The administration of bST to beef heifers at the initiation of a TAI protocol increased plasma concentrations of IGF-1 at TAI; however, failed to enhance follicle diameter, embryo/fetal size, and reduced PR/AI.

To determine the effects of administration of 25 mg of PGF2α 7 d prior to the initiation of the 7-d CO-Synch + controlled internal drug release (CIDR) fixed-time AI (TAI) protocol, 985 Bos taurus beef heifers were enrolled in a completely randomized design at 9 locations from April to July of 2016. Within location, all heifers were randomly assigned to 1 of 2 treatments: 1) CONTROL (n = 496); 100 µg injection of GnRH and a CIDR insert for 7 d [day 7], administration of 25 mg of PGF2α at CIDR removal [day 0], followed by a second injection of GnRH and TAI 54 ± 2 h later; or 2) PRESYNCH (n = 489); same as CONTROL but heifers received an additional injection of 25 mg of PGF2α 7 d prior [day 14] to CIDR insertion. Estrous detection patches were applied to all heifers on day 14 and were evaluated for estrual activity on day 7. Similarly, estrus alert patches were placed on all heifers on day 0 and evaluated for estrual activity at the time of TAI. Pregnancy was diagnosed via transrectal ultrasonography between 35 and 55 d after TAI. The percentage of heifers exhibiting estrus between days 14 and 7 was greater (P < 0.001) for the PRESYNCH (70.1 ± 2.4%) than the CONTROL (41.1 ± 2.3%) treatment, whereas the percentage of heifers exhibiting estrus between day 0 and TAI was greater (P < 0.001) for the CONTROL (55.6 ± 2.4%) than the PRESYNCH (39.7 ± 2.5%) treatment. Estrus response rates differed (P < 0.001) among locations. Pregnancy rates to TAI differed (P = 0.023) among locations; however, they did not differ (P = 0.739) between CONTROL and PRESYNCH treatments (45.4 ± 2.5 vs. 43.2 ± 2.5%, respectively). Final breeding season pregnancy rates did not differ (P = 0.811) between treatments. Therefore, an injection of PGF2α 7 d prior to initiation of the 7-d CO-Synch + CIDR protocol failed to improve pregnancy rates to TAI in replacement beef heifers.

Two experiments were conducted to evaluate the effects of a high concentrate, s.c. PGF2α compared with a conventionally concentrated, i.m. PGF2α in estrus synchronization protocols for heifers. In Exp. 1, 869 Angus-based beef heifers were enrolled at 8 locations. All heifers were exposed to the 7-d CO-Synch + controlled internal drug release (CIDR) estrus synchronization protocol. On day 7 of the protocol heifers received 100 µg of GnRH i.m., and a CIDR insert for 7 d. On day 0, at CIDR removal, estrous detection patches were applied to heifers and, within location, heifers randomly received 1 of 2 PGF2α treatments: 5 mL of Lutalyse i.m. (CONTROL; n = 434) or a 2 mL of Lutalyse HighCon s.c. (HiCON; n = 435). A second GnRH injection was administered at 54 ± 2 h and heifers were fixed-time AI (TAI). Heifers were evaluated for estrous activity at TAI by determining the activation of estrous detection patches. Pregnancy rates to AI (PR/AI) were diagnosed by transrectal ultrasonography between 35 and 55 d after TAI. The percentage of heifers exhibiting estrus between day 0 and TAI did not differ (P = 0.68) between CONTROL and HiCON treatments (47 vs. 46 ± 4%, respectively). Additionally, PR/AI were similar (P = 0.65) between CONTROL and HiCON treatments (46 vs. 45 ± 3%). In Exp. 2, 190 Angus-based beef heifers were enrolled at 2 locations. Heifers were exposed to the melengestrol acetate (MGA)-PGF2α protocol where they were offered 0.5 mg MGA per day from days 1 to 14. On day 33, heifers were randomly assigned to receive CONTROL (n = 95) or HiCON (n = 95) treatment, and estrous detection aids were applied. Heifers were exposed to AI 12 h after detection of estrus. Heifers not detected in estrus at location 1 received a second PGF2α injection 6 d after the initial PGF2α injection and were placed with fertile bulls. Heifers at location 2 that did not express estrus were administered 100 µg of GnRH i.m. and exposed to TAI 96 h after the initial PGF2α injection. Transrectal ultrasonography was used to diagnose PR/AI between 51 and 57 d after the initial PGF2α injection. The percentage of heifers exhibiting estrus during the estrus detection period was similar (P = 0.40) between CONTROL and HiCON treatments (82 vs. 87 ± 4%). Furthermore, PR/AI were similar (P = 0.62) between CONTROL and HiCON treatments (60 vs. 65 ± 5%). In summary, the 2 concentrations and corresponding routes of administration of PGF2α were similar in efficacy at synchronizing estrus in beef heifers.
